I’ve been playing with different defenders on different stages. I used Jessica Jones on the Deadpool challenge, Luke Cage 3* on Strange Sights and Luke Cage 4* on the Orb of Agamotto stage. The counter hasn’t increased at all; it’s stuck at 0. Those are all Defenders, so…bugged?

This seems to be a common bug with these holiday quests. It seems that it should work properly if you complete and claim one of the daily quests first: Problem with Events counter for PVP Holiday Season - #14 by stef_focus - MPQ Bugs and Technical Issues - 505 Go! Official Forums.
Doesn’t work for me. I’m completing daily quests as I go, and I was even able to finish the one asking me to finish the Strange Sights quest “for a true fan” or something like that. But not the Defender’s quest. I’ve gotten other milestones as well. Just not this one.
Today it’s the CHAMPIONS Event that’s not registering for me.,
Same for me, never had this type of issue before the defenders quest.
Oh well, so much for holiday quests. Maybe next year, MPQ.
